I’m surprised this hasn’t been asked, already!
If the show was to tour (a bit unlikely, but IF, remember) and auditions were being held, would you audition, who would you audition to be and what would your song be, if they asked for one from the show?
If it was me:
Of course, I’d audition! (I’m an actor and a Dr. Horrible fan!)
I would be going for Dr. Horrible (he’s got so much character about him)
And my song would probably be either “Everything” or “Brand New Day”.
And if there ever are any auditions, I want to be informed!!!!!! Or the Stun Ray will turn into a Death Ray veeeeeeeeeery quickly!

I'd audition for Captain Hammer. While not quite as tall and good looking as Nathan, I'm pretty buff. Plus, my singing voice fits his better than NPH's.
But if I couldn't land that role (entirely possible since there are many, many better singers than me in this world, country, state, city…apartment building) I'd try for the gay guy who backs up the groupies.
And I'd sing Everybody's a Hero to audition.

I'd have to be penny, obviously.
Well, I have red hair, and I'm a pretty good singer and can sing all the songs.
Although, if they decide a little rewrite and allow me to go for horrible…well who am I to refuse?

I'd be in the pit. Piano, keyboard (not the same as piano, believe you me), trumpet or tenor sax (and no, I can't play all sax — not a transposing issue but an embrasure issue… unless tuba or valved trombone are options, in which case, they'd be options for me, too). My violin is only at the beginning stages. (I still have trouble stringing the darned thing.) My singing has been on a brief 21(+?) year hiatus. I can still sight-read pretty well. My music memorization is pretty good still (though anything else has gotten rather iffy). I haven't really tried improv in over 8 years. Gave it all up for a career in compy wrangling.
Or an extra. I've done that, too… once… only.
Or handling graphic design (only 1 year hiatus, still remember most of the copyright and legal identity requirements and rules, but I'm slowing losing the RAM regarding all that).
